Scientists also learned how the circadian rhythms of the bodies of little mice (and perhaps humans too) are very dependent on the workings of the microbiome. Surprisingly, genes that show no signs of circadian rhythms take over when these microbial rhythms are disrupted too.

These findings were observed when researchers gave mice antibiotics that removed much of their gut bacteria and even when their feeding times were changed.

Circadian rhythms and obesity

This inter-dependence between the gut and the body’s circadian rhythms could play a role behind the scenes in promoting the accumulation of body fat that leads to obesity, according to a recent study conducted at the University of Texas Southwestern and published in the journal, Science.

Based on tests that compared the health of germ-free, conventionally raised and genetically modified mice, scientists learned how the microbiome controls fat uptake and storage by “hacking into” and altering the functions of the circadian clocks within cells that line the gut.

They identified the mechanism by which gut bacteria regulate the composition of body fat and use a chemical (circadian transcription factor NFIL3) to establish a critical molecular link between the microbiota, circadian clock and metabolism, says Dr. Lora Hooper, lead author of the study, according to a press release.

So how does the microbiota hack into the lining of the gut?

A body’s circadian clock senses those day and night cycles, which are linked closely to feeding times, and transmits that information to the gut to turn on and off the metabolism (the uptake of lipids) when necessary.

More specifically, the circadian rhythms of the gut control the expression of NFIL3 and the production of lipids that are governed by this chemical in the intestinal lining.

“So what you have is a really fascinating system where two signals from the environment come in – the microbiome and the day-night changes in light – and converge on the gut lining to regulate how much lipid you take up from your diet and store as fat,” says Dr. Hooper.

Treating IBS-Related Depression With Probiotics

Multi-species probiotics conquer constipation

Can excessive exercise promote leaky gut?

The benefits from regular exercise — from reducing your risks to catastrophic conditions like cancer, type 2 diabetes and cardiovascular disease to improving your brain — are many and varied. In fact, a very popular Lancet study argued the lack of regular exercise could be as deadly to your health as smoking.

But, what happens when you take exercise to the opposite extreme? Overdoing anything often reverses much of the benefits you might’ve achieved in moderation, and exercise is no exception.

Based on a 2015 study published in the Journal of the American College of Cardiology, people who exercised more than four hours a week had virtually the same risk of dying as sedentary folks who rarely exercised.

A pair of very recent scientific reports have also tied excessive exercise to serious health problems that may promote leaky gut, a disorder in which a breakdown in the intestinal wall allows unintended substances — undigested food, toxic waste products, bacteria and viruses — to seep through the intestinal barrier and into the bloodstream.

Extreme exercise may harm your gut after 2 hours

Exercising for two hours at 60 percent of the maximum volume of oxygen an athlete can use (VO2 max) along with stress felt from excess heat pushed patients into an unhealthy state, based on a review of studies featured in Alimentary Pharmacology and Therapeutics.

Lengthy military training may harm the gut too

In this study, military scientists from Norway, the U.S. Army and the Geneva Foundation as well as the Frederick National Laboratory for Cancer Research tracked the health of 73 Norwegian Army soldiers training in cross country skiing for four days. During that time, soldiers skied 31 miles while carrying 99-pound backpacks.

Researchers collected blood and fecal samples before and after the exercise. Before giving urine samples on the first and third days of training, soldiers drank a mix of water and sucralose and mannitol (an artificial sugar and a sugar alcohol, respectively) to detect signs of leaky gut.

By the end of the training period, the collective gut health of these soldiers as well as the composition of substances in blood and stool samples taken from them changed significantly and for worse. The excretion of sucralose rose greatly too, indicating an increase in leaky gut.

Despite the good probiotics do, some scientists believe they may not do much to protect gut permeability. However, a 2012 report from the Journal of the International Society of Sports Nutrition that tracked the health of 22 trained athletes taking multi-species probiotics proved otherwise.

Mangos may protect your gut from high-fat diets

Scientists at Oklahoma State University and North Carolina State University compared the health of 60 male mice, based on feeding them high-fat diets supplemented with or without mangos and in varying amounts (1 or 10 percent).

Animals fed a high-fat diet plus 10 percent in mangos — equal to humans eating 1.5 cups of this delicious fruit — retained more of their gut bacteria than those fed lower amounts.

Also, the addition of mangos to the diets of mice resulted in their guts containing more gut bacteria, specifically Bifidobacteria and Akkermansia (bacteria found in lower amounts in obese animals), and improved production of short-chain fatty acids (SFCAs), beneficial compounds that have anti-inflammatory properties.

“Fiber and other bioactive compounds in plant-based foods are suggested to prevent gut dysbiosis caused by a high-fat diet,” said Edralin A. Lucas, Ph.D., professor of nutritional sciences at Oklahoma State and lead researcher of the study, according to a press release.

“The results of this animal study showed that adding mango to the diet may help maintain and regulate gut health and levels of beneficial bacteria levels.”

What’s ahead

Researchers caution more work needs to be done to confirm the benefits of eating mangos on the human gut once and for all, especially for those whose diets aren’t the healthiest.

Could poor gut health trigger Alzheimer’s?

Before those transplants, germ-free mice had significantly smaller amounts of beta-amyloid plaque, protein fragments that build up between neurons in the brain. After the transplants, even those “clean” animals were vulnerable to the growth of brain-killing beta-amyloid plaque.

Typically, the healthy brain breaks down those fragments and sheds them. As beta-amyloid plaque and neurofibrillary tangles accumulate in the brain, however, the symptoms of Alzheimer’s begin to present themselves.

“Our study is unique as it shows a direct causal link between gut bacteria and Alzheimer’s disease,” says Dr. Frida Fåk Hållenius, according to a press release. “The results mean that we can now begin researchers ways to prevent the disease and delay the onset.”

A balanced gut may tame autism

Is medicine better able to spot signs of autism spectrum disorder (ASD) or is the incidence of this challenging developmental disability actually growing?

It’s a question without a concrete answer…

Although the numbers remained stable from 2010 to 2012 according to the most recent CDC stats (2012), autism affects 1 in 68 children, and it’s some 450 percent more common among boys than girls.

Despite this lack of clarity, fortunately, researchers have identified a pretty common problem kids across the spectrum share: Gastrointestinal challenges that could be related to taking too many antibiotics, thus reducing their microbial diversity too early in their young lives.

A pair of recent studies highlighted the range of gut-based treatments that may soon tame the effects of autism.

 

Going the fecal transplant route

A research team from Arizona State University, Northern Arizona University and Ohio State University experienced success treating a small group of 18 kids using fecal transplants, according to a study appearing in Microbiome.

Over 10 weeks, this young group of patients, ranging in age from 7-16, were treated with a bowel cleanse and two weeks of antibiotics, followed by daily fecal transplants for eight weeks.

The treatment regimen was effective, as it improved gastrointestinal symptoms by some 80 percent along with gut microbial diversity and calmed autistic behaviors by around 25 percent. And, that gut diversity held after treatments ended too.

But, there’s a couple of caveats to consider about this study too. For starters, there was no side-by-side placebo trial that would demonstrate if doing nothing made a difference too. That’s not a big deal.

The larger concerns, however, are the unintended consequences of having a fecal transplant in the first place, like receiving gut bacteria from someone who may be reasonably healthy but overweight then acquiring that same health problem.

Also, these risks can be especially tricky when patients attempt to take matters into their own hands by trying to replicate these treatments, a real problem researchers have acknowledged.

 

A dietary/probiotic solution?

The lack of diversity in the gut was important to another study, this time with mice bred to exhibit autism-like behaviors, according to a 2016 report by Baylor College of Medicine researchers appearing Cell.

Based on sequencing long strands of DNA, the offspring of mice moms fed high-fat diets were severely lacking in Lactobacillus reuteri in their guts by a factor of nine.

Restoring that balance lessened some autistic behaviors and boosted the production of oxytocin, a hormone that acts like a neurotransmitter in the brain and a facilitator of bonding

Researchers speculate this genus of Lactobacillus may provide a basis for treating human neurodevelopmental issues in the form of a probiotic.

Could your gut be training your body to yo-yo diet?

Losing weight isn’t easy. It takes a lot of consistent effort in many areas — exercise, food choices, portion control, sleep, self-esteem are just a few — to do it the safe and right way.

Sadly, life often gets in the way and not every weight loss effort goes as planned. Sometimes, this can lead to weight cycling, better known as yo-yo dieting.

Although there’s no general consensus among medical experts whether repeatedly losing and regaining weight is bad, there are health consequences associated with yo-yo dieting, like coronary issues, extra stress and a slower metabolism.

A recent series of tests by a team of Israeli researchers pinpointed a potential cause for yo-yo dieting in a study appearing in Nature: A gut microbiome that changes when weight is lost, then exposed to high-fat foods again.

 

The experiments

As scientists studied mice, they discovered an important constant with yo-yo dieting: After one cycle of gaining and losing weight, every bodily system in their test subjects reverted to normal except for their microbiomes. For some six months after their weight loss, mice retained an “obese” microbiome.

“This persistent microbiome accelerated the regaining of weight when the mice were put back on a high-calorie diet or ate regular food in excessive amounts,” said lead researcher Dr. Eran Elinav of the Weizman Institute of Science in a press release.

No surprise, when researchers transplanted gut bacteria from obese mice into germ-free mice, they began to gain weight too when fed high-fat foods.

It was only when scientists bombarded obese mice with broad-spectrum antibiotics or gave them fecal samples from mice that had never been obese that the cycle stopped.

Those treatments may work for mice, but for humans, antibiotics have been a known enemy of gut health for a very long time and fecal transplants have unintended consequences that may do more harm than good.

However, scientists identified a pair of flavonoids, a diverse family of natural chemicals found in nearly all fruits and vegetables, that were in short supply among obese mice that would improve fat-burning.

When mice were fed flavonoids in their drinking water, their little bodies readjusted and didn’t experience accelerated weight gains, even when fed high-calorie diets.

 

Targeting the gut

Whether extra flavonoids will work on the guts of humans to prevent yo-yo weight gains is anyone’s guess. However, there’s one critical aspect of gut health that the Israeli study didn’t investigate.

A diverse gut protects your health during immunotherapy treatments for cancer

For many cancer patients, undergoing chemotherapy or radiation are often a necessity, but they come with lots of risks depending on the severity and length of treatments.

Rather than bombarding tumors with chemo and radiation, however, some patients and their teams of doctors are choosing other cancer-fighting approaches like immunotherapy that work far differently.

Immunotherapy focuses on treating your body’s immune system to fight cancer either by supercharging a patient’s immune system or teaching his/her body how to spot cancer cells and eradicate them. Also, in some cases, immunotherapy can aid in a cancer patient’s recovery long after treatments have ended.

But not everyone responds well to immunotherapy, which has researchers scrambling for answers.

Over the years, cancer researchers have learned how good gut health plays a critical role in protecting cancer patients during chemo treatments.

A diverse gut microbiome may also be very important in how well the human body handles certain forms of immunotherapy, according to a study presented at a recent symposium sponsored by American Society of Clinical Oncology.

A team of researchers, led by senior study author Dr. Jennifer Wargo from the University of Texas MD Anderson Cancer Center, studied the connections between a healthy gut and the benefits of immunotherapy by examining fecal and oral bacteria samples taken from more than 200 patients fighting metastatic melanoma, an advanced form of skin cancer.

Ninety-three patients received an anti-PD1 immune drug that blocked a pathway protecting tumor cells from a patient’s immune system equipped to fight it.

From that smaller group, scientists studied fecal samples provided by 30 patients who responded to immunotherapy and 13 more who didn’t.

No surprise, patients who responded to the anti-PD1 drug had greater diversity of gut bacteria and for a specific type of bacteria (Ruminococcaceae). Plus, an examination of their tumors uncovered a greater number of cancer-fighting immune system cells (CD8+T).

On the other hand, patients whose bodies didn’t react to immunotherapy drugs had much lower gut diversity and one specific family of gut bacteria (Bacteriodales).

“Meanwhile, we need concerted research efforts to better understand how the microbiome may influence immune responses, as well as an in depth view on how we can tweak the microbiome so that more patients can benefit from immunotherapy,” said Dr. Wargo, an associate professor of genomic medicine and surgical oncology, according to a press release.

Some of that tweaking may come from changing a patient’s dietary habits or boosting the diversity of their gut by recommending a probiotic, scientists said.

IBS and the gut-brain axis

Curious about the effect irritable bowel syndrome (IBS) has on behavior and intestinal health, researchers from McMaster University and the University of Waterloo took an unconventional approach: Transplanting fecal samples from eight patients suffering from IBS with diarrhea for at least two years and five healthy people into germ-free mice.

Three weeks later, compared to mice that received healthy samples, animals that were given IBS-laced transplants experienced increased gut permeability, low-grade inflammation and faster gastrointestinal transit (how long it takes food to travel from the stomach and through the intestine).

Then, scientists tested anxiety-associated behaviors by measuring the time mice spent in the dark and how long it took them to step down from a platform to explore their environments, according to The Scientist.

Mice that were given fecal samples from human IBS patients who reported anxieties experienced similar emotional difficulties, compared to animals given fecal samples from healthy patients and those with IBS who reported no problems with anxieties.

Probiotics to the rescue

Researcher Dr. Giada De Palma called these findings a landmark “because it moves the field beyond a simple association and toward evidence that changes in the microbiota impact both intestinal and behavioral responses in IBS.”

These results also offer more evidence that human gut health may play a larger role in the range of brain disorders ranging from the emotional (mood or anxieties) to more serious problems, like multiple sclerosis, Parkinson’s disease and autism, according to a press release.

The good news: Scientists believe treatments such as probiotics and prebiotics could be beneficial in treating, not only the physical aspects of IBS but the behavioral issues associated with it too.
